ACQUITY BEH Glycan 1.7um Method Validation Kit includes three batches of chromatographic media [derived from different base particles] to judge the quality, reliability, and consistency of your chromatographic method. Each batch of selected ACQUITY BEH Glycan 1.7um chemistry is specifically QC tested with a mixture of 2-AB labeled glycans to help ensure consistent performance.

ACQUITY UPLC BEH Amide Columns

ACQUITY BEH Amide columns retain extremely polar compounds, including carbohydrates and sugars. Based on BEH technology the ACQUITY Amide columns offer a chemically stable, trifunctionally bonded amide phase that is stable from pH 2 to 12 to enable the separation of polar analytes that span a wide range of polarity, structural moiety and pKa.

Since 2003, Waters has developed innovative stationary phases  for Hydrophilic Interaction Chromatography [HILIC] to overcome the  challenge of retaining and separating extremely polar compounds. Based on Waters novel ethylene bridged hybrid [BEH] particle technology, New ACQUITY UPLC® BEH Amide columns utilize a chemically stable, trifunctionally-bonded amide phase, enabling a new dimension in stability and versatility for HILIC separations.

Designed to retain polar analytes and metabolites that are too polar to retain by reversed-phase [RP] chromatography, ACQUITY UPLC BEH Amide columns facilitate the use of a wide range of mobile phase pH [2 -11] to facilitate the exceptional retention of polar analytes spanning a wide range in polarity, structural moiety and pKa.

In addition to enhanced retention of polar compounds, ACQUITY UPLC BEH Amide columns provide increased mass spectrometry response, direct compatibility with sample preparation eluates [PPT, LLE and SPE] as well as orthogonal selectivity compared to reversed-phase materials, making HILIC an attractive alternative separation technique.

BEH (Ethylene Bridged Hybrid) Technology

In 1999, Waters launched the XTerra family of HPLC columns featuring first generation hybrid particle technology (HPT). HPT enabled XTerra columns to become one of the most successful column products in the history of Waters. In HPT, the best properties of inorganic (silica) and organic (polymeric) packings are combined to produce a material that has superior mechanical strength, efficiency, high pH stability and peak shape for bases. The first generation methyl hybrid particles of XTerra columns did not possess the mechanical strength or efficiency necessary to fully realize the potential speed, sensitivity and resolution capabilities of UPLC technology. Therefore, a new pressure-tolerant particle needed to be created. A new, second generation hybrid material was developed which utilizes an ethylene bridged hybrid (BEH) structure. Compared to the first generation methyl hybrid particle of XTerra columns, the BEH particle of ACQUITY UPLC BEH columns exhibits improved efficiency, strength and pH range. BEH Technology™ is a key enabler behind the speed, sensitivity and resolution of UPLC separations.

ACQUITY UPLC columns

ACQUITY UPLC columns are the most technologically advanced LC columns ever created and are the only columns designed, tested and certified to withstand the pressures of UPLC Technology. Based upon the highly efficient 1.7 µm Bridged Ethyl Hybrid (BEH) particles and 1.8 µm High Strength Silica (HSS) particles, these columns are the first and only columns that allow separation scientists to achieve maximum speed, sensitivity and resolution without compromise. The particles and chemistries of the ACQUITY UPLC column family were carefully chosen to produce the ideal combination of ultra-performance efficiencies, wide pH range and complementary selectivities. ACQUITY UPLC BEH columns and ACQUITY UPLC HSS columns are the only columns recommended for use with ACQUITY UPLC Systems.

There is more to creating a UPLC particle than synthesizing a small particle. Many HPLC particles do not possess the mechanical stability and structural integrity to withstand UPLC operating pressures (e.g., 15000 psi/1000 bar). Why is pressure tolerance important? In order to realize the efficiency gains of sub-2 μm particles, the ability to routinely operate at higher linear velocities (e.g., higher flow rates) is required. These higher linear velocities combined with small, sub-2 μm particles result in higher operating backpressures. Waters has created two highly efficient, pressure-tolerant UPLC particles: the 1.7 μm Ethylene Bridged Hybrid (BEH) particle and the 1.8 μm High Strength Silica (HSS) particle.

The first ACQUITY UPLC particle created was the 1.7 μm Ethylene Bridged Hybrid (BEH) particle. This second generation hybrid particle is one of the key enablers behind UPLC technology and is available in five column chemistries: C18, C8, Shield RP18, Phenyl and HILIC. Because this is a hybrid particle, a wider usable pH range (up to pH 1-12) makes method development faster and easier. BEH particles are also available in HPLC particle sizes (2.5, 3.5, 5 and 10 μm) in the XBridge family of HPLC columns, thus allowing seamless transfer between HPLC and UPLC separations.
