Based upon the highly successful BEH particle technology, the ACQUITY UPLC Protein BEH SEC Column, 200 Å, 1.7 µm, 4.6 mm X 150 mm represents the first commercially available sub-2-µm SEC chemistry. Fully optimized for the analysis of proteins and their aggregates with molecular weights ranging from 10000 to 450,000 Daltons, the total system solution will routinely deliver fast, accurate aggregate determinations up to 10 times quicker that traditional HPLC-based SEC methods. Note: The same column is also available with the BEH200 SEC Protein Standard at no additional cost. (PN: 176003904) Also note that for optimal SEC separation performance, this column is designed for use in UPLC SEC applications with an appropriate low LC system volume.

BEH (Ethylene Bridged Hybrid) Technology
Ethylene Bridged Hybrid (BEH) particle technology ensures maximum column performance and longer column lifetimes under all chromatographic conditions. The hybrid particle technology offers many advantages over conventional silica-based particles, including the ability to control silanol activity for better reproducibility, peak shape, and efficiency. The BEH particle is created from two high purity monomers: tetraethoxysilane (TEOS) and bis(triethoxysilyl) ethane (BTEE), which results in highly stable, pH resistant, and mechanically strong particles that set the performance standard for method development. BEH particle technology allows for seamless transfer from analytical to preparative separations.

SEC is a powerful technique for aggregate and fragment analysis in the research, development, and manufacturing of biotherapeutic proteins, such as insulin and monoclonal antibodies (mAb). With SEC, also known as gel filtration, you can separate proteins based on their size (hydrodynamic radii versus absolute molecular weight) in solution, with larger sized species eluting before smaller proteins.
The pore size and volume of the SEC particles used in the packed column drives this isocratic based technique, so the column you choose matters. As the molecular weight of the proteins of interest increases, SEC columns with comparatively larger pore size particles are selected.
