Whether you are looking into the metabolomics of a disease through changes in TCA cycle carboxylic acids, or trying to analyze phosphoric or even sulfuric acids, attaining good chromatography for organic acids (OA), can be quite a challenge. This is true for several reasons. First, OAs are polar making them difficult to retain on traditional reversed phase columns without ion-pairing or derivatization, which further complicates analysis and data interpretation. Second, the presence of multiple pKa values can lead to variations in the charge states of your analytes, which may cause poor peak shapes including tailing, shouldering and even split peaks. Additionally, OAs are typically poor chromophores, making them difficult to analyze by UV-Vis detection, and CAD does not work well with gradient elutions. Recognizing these challenges, we developed them most complete solution for OA analysis, with a novel stationary phase called Intrada Organic Acid. This proprietary multi-mode column optimizes the use of multiple stationary phase structures specifically targeting OAs, delivering the most complete LC-MS-based solution on the market today.

Importance of sample and solvent preparation
When using Intrada Organic Acid, special care should be given to preparation of the sample solvent, especially pH. This is particularly important when analyzing compounds with low pKa values or polyanionic species, where preparation of sample solvents with different pHs can lead to inconsistent results. Proper preparation of the sample itself is also important, to increase column performance and lifetime, paying particular attention to the removal of large polymers such as proteins.
Additional Considerations
- The Intrada Organic Acid column should only be used with LC-MS. This product is not recommended for UV, Electrical Conductivity Detector or ELSD detection, in order to avoid peak identification failure.
- While this product is ideal for most organic acids, we cannot guarantee that it will work for all of them, particularly for some isomers.
- Detection sensitivity is highly dependent on the structure of the compound as well as the capabilities and setup of the MS instrument, especially the drying gas temperature.
- The sample matrix can have a big impact on peak shape, so sample preparation and sample solvent should be optimized to improve peak shape.
- The guard cartridges for this product are not metal-free, so we do not recommend using them for phosphoric, chelating or ion-suppressing compounds.

This metal-free column is an Imtakt original design. The entire internal surface of these columns are made with PEEK material, including the frits, and is therefore extremely inert with respect to chelating compounds. The outer portion of these columns are made from stainless steel (SS316) making them suitable for extremely high pressures, and are UHPLC compatible.
Phosphoric compounds can interact with the exposed metal surfaces within the column causing peak tailing, as seen in the figure to the left.
Imtakt's metal-free columns are designed to specifically improve these poor peak shapes by decreasing unwanted secondary chelating effects.
Improving peak shape of chelating or phosphoric compounds
Chelating and phosphoric compounds often have difficulty with poor peak shape and/or sensitivity due to undesired interaction between metal parts in the column and the structure of these compounds. Imtakt's UHPLC compatible metal-free columns, which are an Imtakt original design, will improve these
problems and are available in a wide range of stationary phases.
