The RSpak NN-814 and JJ-50 2D combine the reversed phase (RP) mode with ion exchange chromatography (IEC)

RSpak NN-814 packed with polyhydroxymethacrylate gel is a column for reversed phase mode. A small number of sulfo groups (counter ion H+) bonded to the packings, however, characterize NN-814. When a neutral or acidic eluent is used, acidic, neutral and basic substances in principle elute in this order. Taking this advantage, NN-814 enables complicated components such as permanent wave solution to be analyzed by simple isocratic elution with good reproducibility.In NN-814, ion exclusion, ion exchange and reversed phase modes have effect on separation. Here, acidic substances such as TGA and DTGA elute first as they are affected mainly by the ion exclusion mode. In contrast, basic substances such as ammonia and monoethanolamine elute later as they are affected mainly by the ion exchange mode. Other components are affected mainly by the reversed phase mode.

RSpak NN-814 Features

Eluents

Following eluents can be used: (1) Buffer, (2) Buffer + salt, (3) Buffer + polar organic solvent, (4) Buffer + salt + polar organic solvent. The maximum concentration of organic solvents, methanol, ethanol and acetonitrile is 50%. 

Separation of the sample can be controlled by changing eluent:

Water / polar organic solvent

  • The elution time of substances which have long alkyl chain or benzene ring is rather long.
  • The elution time of such substances can be shorten by raising the concentration of polar organic solvent.

pH

  • In the case of basic substances, the lower the eluent pH value, the faster the elution time.
  • The pH range of eluent should be 2.0 to 12.0. In the case of eluent containing chlorine ions, the pH should be 4.0 or higher.
  • Strong acids, strong bases and perchloric acid should not be used.

Salt concentration

  • In case of basic substances, the higher the salt concentration of the eluent, the faster the elution time.
  • The maximum concentration of salt is 0.5 M

Column temperature

  • The elution time of ionic substances changes according to the temperature.

Cleaning/Regeneration

Inject 20 to 50 μL of 2 N nitric acid aqueous solution two or three times.
