Gel Filtration Chromatography
TSKgel UltraSW Aggregate Column, 3 μm, ID 7.8 mm x Length 30 cm
TSKgel UltraSW columns, represent the fourth generation of high performance gel filtration columns that have become synonymous with analyzing protein molecular weights in the field of biotechnology. It is based on even smaller silica beads than TSKgel SuperSW 3000 with slightly larger pore size. The separation range of TSKgel UltraSW Aggregate therefore covers higher molecular weights than TSKgel SuperSW mAb HR. It provides a very good separation of larger proteins or antibody dimers and higher aggregates.
Particle Characteristics TSKgel UltraSW-type columns contain smaller particles than SuperSW-type columns: 3 micron versus 4 micron. The surface has been shielded from interacting with proteins by derivatization with ligands containing diol functional groups.
Calibration Curves The figure to the right shows the calibration curves for globular proteins for the TSKgel UltraSW Aggregate column in comparison to the two TSKgel SuperSW-mAb columns that are also dedicated to antibody analysis, TSKgel SuperSW mAb HR & TSKgel SuperSW HTP.
HPLC System Requirements It is important to employ an HPLC system that is optimized with regards to extra-column bandbroadening to take full advantage of the high column efficiency that can be obtained on the TSKgel UltraSW columns. See SEC brochure for recommendations.
