ACQUITY Premier Protein SEC Column, 250Å, 1.7 µm, 4.6 x 300 mm Method Validation Kit Columns help scientists obtain reliable protein size variant separations (10,000 to approx 650,000 Daltons) via the synergistic coupling of Waters MaxPeak Premier High Performance Surfaces and novel BEH-PEO Particle technologies. These advancements in SEC column hardware and SEC particle technology work to minimize non-desired secondary ionic or hydrophobic interactions between proteins and the column to minimize costly method development work by allowing chromatographers to use a “generic” or “platform-type” method for many samples. Each batch is specifically QC tested using Waters mAb Size Variant Standard (p/n: 186009429) to help ensure consistent performance for challenging size-based protein separations.

SEC is a powerful technique for aggregate and fragment analysis in the research, development, and manufacturing of biotherapeutic proteins, such as insulin and monoclonal antibodies (mAb). With SEC, also known as gel filtration, you can separate proteins based on their size (hydrodynamic radii versus absolute molecular weight) in solution, with larger sized species eluting before smaller proteins.

The pore size and volume of the SEC particles used in the packed column drives this isocratic based technique, so the column you choose matters. As the molecular weight of the proteins of interest increases, SEC columns with comparatively larger pore size particles are selected.
