Testing of semi-solid dosage forms such as creams, gels, and ointments has become more common since the introduction of USP chapter <1724>. While use of the Franz Diffusion Cell (also known as the vertical diffusion cell) is most common, it often requires the purchase of new instrumentation for labs that typically rely on dissolution testing. The Immersion Cell offers an alternative means of testing by using existing dissolution testers.

In-Vitro Release Testing (IVRT) as discussed in USP <1724> is a means of comparing two semi-solid dosages. The test is run under conditions so that the API releases at the maximum rate and no impediment through sink conditions or a restrictive membrane is allowed. The test can be used as a means of ensuring batch to batch uniformity or comparing innovator and generic products. For more information in IVRT, refer to USP <1724>, or the FDA Guidance for Industry Nonsterile Semisolid Dosage Forms Scale-Up and Postapproval Changes (FDA, 1997).

The Immersion Cell is primarily a means of release rate testing for semi-solid products. The user places a membrane into the cell then fills the cavity with the dosage. After the cavity is filled, the user will place the cell into a flat bottomed dissolution vessel where the drug passes through the membrane and is released into receptor media.

Immersion Cells typically offer the following advantages over Franz Diffusion Cell systems:

  • Can use existing dissolution testers rather than requiring the purchase and qualification of new instruments.
  • Cells are surrounded by a water bath rather than water jacket which provides better temperature control and stability making it ideal for temperature sensitive products.
  • Easier to use as staff familiar with dissolution testing do not have to learn new techniques which means usable results can be obtained quickly.
